Optimization of the Ussing chamber setup with excised rat intestinal segments for dissolution/permeation experiments of poorly soluble drugs.

2016

AbstractContext: Prediction of the in vivo absorption of poorly soluble drugs may require simultaneous dissolution/permeation experiments. In vivo predictive media have been modified for permeation experiments with Caco-2 cells, but not for excised rat intestinal segments. Objective: The present study aimed at improving the setup of dissolution/permeation experiments with excised rat intestinal segments by assessing suitable donor and receiver media. Methods: The regional compatibility of rat intestine in Ussing chambers with modified Fasted and Fed State Simulated Intestinal Fluids (Fa/FeSSIFmod) as donor media was evaluated via several parameters that reflect the viability of the excised in…

Formulation strategy towards minimizing viscosity mediated negative food effect on disintegration and dissolution of immediate release tablets.

2017

Food induced viscosity can delay disintegration and subsequent release of API from solid dosage form which may lead to severe reduction in the bioavailability of BCS type III compounds. Formulations of such tablets need to be optimized in view of this postprandial viscosity factor. In this study, three super disintegrants, croscarmellose sodium (CCS), cross-linked polyvinylpolypyrrolidone (CPD), and sodium starch glycolate (SSG) were assessed for their efficiency under simulated fed state. Tablets containing these disintegrants were compressed at 10 and 30 KN, while taking lactose as a soluble filler. In addition to other compendial tests, disintegration force of these formulations was meas…

982 Maternal Age, History of Migration and Incidence of Infantile Hypertrophic Pyloric Stenosis in Germany in 2000-2008

2012

Background The incidence of infantile hypertrophic pyloric stenosis (IHPS) is highly variable over time and geographic regions. A decline in IHPS incidence was recently reported in Sweden, the US, Denmark, and Scotland and in Germany. Aim In further evaluation of our previous epidemiological data, we collected data on maternal age and history of migration in mothers from the regional administrations. We examined correlations between these factors and IHPS incidence. Methods Data were extracted from the public report of Health (Gesundheitsberichterstattung des Bundes) and population data from federal state governments. We collected the numbers of IHPS (International Statistical Classificatio…

Improvement of trospium-specific absorption models for fasted and fed states in humans

2014

The purpose of this study was to mechanistically interpret the oral absorption pattern of trospium in fasted and fed states by means of gastrointestinal simulation technology. A drug absorption model was built on the basis of experimental data. According to the generated model, low permeability across the intestinal epithelium, delayed gastric emptying time and a prolonged residence time in the small intestine are the key factors governing trospium absorption in the fasted state. Furthermore, in silico modelling provided a plausible explanation of the pronounced reduction in the oral bioavailability of trospium when administered with food. The simulation results support the decreased dissol…

Usutu virus activity in Austria, 2001–2002

2003

Usutu virus (USUV), a member of the mosquito-borne clade within the Flaviviridae family, was responsible for avian mortality in Austria in 2001. In 2002, the virus continued to kill birds, predominantly blackbirds. High numbers of avian deaths were recorded within the city of Vienna and in surrounding districts of the federal state of Lower Austria, while single die-offs were noticed in the federal states of Styria and Burgenland. A total of 72 birds were submitted for laboratory examination, 30 of which tested positive for USUV by immunohistochemistry and/or polymerase chain reaction. Laboratory-confirmed cases of USUV infection originated from the federal states of Vienna and Lower Austri…

Niemiecka samorządność terytorialna. Bilans pozjednoczeniowego trzydziestolecia przemian ustrojowo-strukturalnych

2020

Local government is one of the most important institutions of a democratic rule of law in Germany. Its position and role was basically shaped immediately after the end of World War II. The purpose of this article is an attempt to present and evaluate the systemic and structural changes that have taken place in German local government within 30 years of the post-unification period. The hypothesis of this study is as follows: The political and structural changes of German local government in the last 30 years have been directed towards making the activities of its bodies and institutions more effective as well as strengthening and deepening democracy, influencing the authorities in power, rat…

Exploring gastric emptying rate in minipigs: Effect of food type and pre-dosing of metoclopramide.

2018

The present study investigated the gastric emptying rate in Gottingen minipigs pre- and post-prandial and evaluated the effect of metoclopramide on the same parameter, using paracetamol as an absorption marker. The pharmacokinetic evaluation of the obtained plasma concentration data for paracetamol demonstrated that the fastest gastric emptying rate was observed in the animals that were allowed access to normal pig food. There was no significant difference in the stomach emptying rate observed between fasted and fed minipigs, when fed either with a FDA standard breakfast or a nutritional energy drink. Pre-dosing minipigs with metoclopramide (0.2 or 0.4 mg/kg) did not demonstrate any effect …

Survey of root canal irrigating solutions used in dental practices within Germany.

2014

Aim Irrigant usage information in relation to years of professional experience was collected from general dentists in different German federal states by means of a questionnaire. Methodology A short survey concerning endodontic treatment and use of irrigants was mailed to 4240 dentists or delivered to 3720 dental offices as an extra page in a journal in eight German states. Detailed information concerning the most frequently used irrigants, their concentrations, the spectrum of disinfectants used in root canal treatment concepts for vital and nonvital pulps, main topics of continuing education as well as years of professional experience was collected. The statistical analysis of differences…

German self-governance. Thirty years of constitutional and structural changes in reunified Germany

2022

Local government is one of the most important institutions of a democracy based on the nile of law in Germany. Its position and role were shaped immediately after the end of World War II. This paper aims to present and evaluate tire constitutional and structural changes that have taken place in German local government within the 30years of the post-unification period. The present study is based on the hypothesis that the constitutional and structural changes of German local government in the past 30years have aimed to make its bodies and institutions Work more effectively as well as to strengthen and deepen democracy, thus increasing its impact on the federal government. These changes have …
